Isolasi dan Identifikasi Senyawa Antioksidan Daun Lai (Durio kutejensis (Hassk.) Becc.) dengan Metode DPPH (2,2-Difenil-1-Pikrilhidrazil)

Abstract

Antioxidants are compounds that can counteract or reduce the negative effects of free radicals in the body, one of the plants that have the potential as an antioxidant is lai (Durio Kutejensis (Hassk.) Becc.) where it has been reported that the ethanolic extract of lai leaves has very strong antioxidant activity. The purpose of this study was to isolate the active compounds contained in lai leaves that have antioxidant activity. The isolation stages went through several processes, namely maceration with ethanol solvent, fractionation with liquid-liquid extraction method, separation by vacuum liquid chromatography (KCV) method, and purification by preparative TLC to obtain isolate A. The results of each isolation stage were tested antioxidant activity was using this method DPPH (2,2-diphenyl-1- picrylhydrazyl) using a UV-Vis spectrophotometer. The antioxidant activity of each isolation step is indicated by IC50 (Inhibitor Concentration). The results showed that the IC50 of the ethanolic extract of lai leaves was 9.539 ppm, the IC50 of the water fraction was 13.43 ppm, the IC50 of the ethyl acetate fraction was 12,571 ppm, and the IC50 of the n-hexane fraction was 13,352. ppm and IC50 isolate A was 251.55 ppm. The active compound isolate A that plays a role in antioxidant activity in lai leaves is thought to be a phenolic compound
