Obesity is correlated with serious health problems such as stroke, heart and vascular disease, and malignancy. One of the triggers for obesity is high fat and high carbohydrates foods which increase Free Fatty Acid (FFA) and Reactive Oxygen Species (ROS), which trigger oxidative stress and chronic inflammation, which play a role in various degenerative diseases. Objective: This research wants to explore the potential and determine the effective dose of a combination of ethanol extract of turmeric rhizome (Curcuma domestica Val.) and gotu kola leaves (Centella asiatica (L) Urb.) on inflammation (represented by Tumor Necrosis Factor Alpha or TNF-α levels). Methods: This research used an in vivo experimental preventive method using 35 mice which were divided into normal group, induction group, comparison group curcumin 1.8 mg/kgBW, test group 1 ERK 200 mg/kgBW, test group 2 EDP 200 mg/kgBW, test group 3 ERKDP 200 : 100 mg/kgBW and test group 4 ERKDP 100 : 200 mg/kgBW. TNF-α levels were measured using an ELISA kit on the 60th day. Results: The study revealed a decrease in TNF-α levels in all test groups namely ERK 200 mg/kg BW, EDP 200 mg/kgBW, and the combination of ERKDP dose 200 : 100 mg/kgBW. Conclusion: The combination of ERKDP dose 200 : 100 mg/kgBW is the most effective in suppressing TNF-α levels in mice.
