Abstract

Consumption of foods high in carbohydrates and fats can cause obesity and increase lipid peroxidation,
which is the process of fat oxidation that produces excess free radicals. These free radicals can damage
body cells and trigger increased levels of malondialdehyde (MDA), which is a byproduct of lipid
peroxidation and a marker of oxidative stress. Increased MDA is associated with cell damage,
inflammation, and increased risk of metabolic diseases, including insulin resistance, diabetes, and
cardiovascular disease. The purpose of this study was to determine the activity of kate mas leaf extract
(Euphorbia heterophylla L.) on MDA levels in obese mice. The research method used 36male white
Wistar rats which were divided into 6 groups, namely the normal group, induction, comparison (orlistat),
and test groups with doses of 50 mg/Kg BW, 100 mg/Kg BW, and 200 mg/Kg BW. The mice were
induced with high-carbohydrate and fat feed for 6 0 days. The parameters observed were MDA levels
in the blood and liver using a UV-Vis spectrophotometer. The results showed that MDA levels in the
liver were higher than MDA levels in the blood. MDA levels in the blood of the normal group were 17.56
± 4.86 nmol / ml, the induction group 54.43 ± 6.61 nmol / ml, the comparison group 24.75 ± 5.17 nmol
/ ml, the 50 mg dose test 24.44 ± 3.62 nmol / ml, the 100 mg dose test 27.14 ± 5.01 nmol / ml and the
200 mg dose test 28.95 ± 6.28 nmol / ml. While the MDA levels in the liver of the normal group were
24.37 ± 4.38 nmol / ml, the induction group 56.48 ± 6.10 nmol / ml, the comparison group 25.09 ± 3.54
nmol / ml, the 50 mg dose test 26.13 ± 3.21 nmol / ml, the 100 mg dose test 36.61 ± 5.30 nmol / ml and
the 200 mg dose test 34.23 ± 1.48 nmol / ml. The level of ethanol extract of golden kate leaves that can
reduce MDA levels most effectively is a dose of 50 mg / Kg BW. The results of statistical tests showed
that the induction group was significantly different from all groups. The conclusion of this study shows
that the ethanol extract of golden kate leaves has activity in reducing MDA levels in obese mice with an
effective dose of 50 mg / Kg BW.
