The ramania plant (Bouea macrophylla Griffith) is a species of the Anacardiaceae tribe, with different names. Ramania plants (Bouea macrophylla Griffith) are still very limited in their use, namely only as fruits. Ramania leaves (Bouea macrophylla Griffith) empirically have the potential to control the wound healing process. One of the bacteria that can be inhibited by ramania leaves (Bouea macrophylla Griffith) is Staphylococcus aureus. The aim is to determine the results of phytochemical screening of 96% ethanol maceration extract of Ramania leaves (Bouea macrophylla Griffith) and the diameter of the inhibition zone of 96% ethanol extract of Ramania leaves (Bouea macrophylla Griffith) against Staphylococcus aureus bacteria by disc method. 96% ethanol extract of ramania leaves (Bouea macrophylla Griffith) by maceration extraction method with 96% ethanol solvent and extract yield of 8.54%. The 96% ethanol extract of Ramania (Bouea macrophylla Griffith) leaves positively contains alkoloids, flavonoids, streoids, saponins and tannins. Antibacterial activity test was performed with 4 concentration series, i.e. 5%; 10%; 15% and 20%, positive control chloramphenicol 30μg/disc, and negative control DMSO 0.1%. The results of the 96% ethanol ramania leaf (Bouea macrophylla Griffith) antibacterial activity test showed that the 5%, 10%, and 15% concentrations were included in the weak category, with inhibition zones of 1.925 mm, 3.355 mm, and 4.737 mm, respectively, while the 20% concentration was included in the moderate category with an inhibition zone of 5.775 mm. From the results of the study can show that the ethanol extract of 96% ramania leaves (Bouea macrophylla Griffith) has antibacterial activity against Staphylococcus aureus bacteria.
